- 以洛巴伊大口蘑为试材,采用袋栽方法,测定了洛巴伊大口蘑在棉籽壳培养基上生长期间栽培基质的降解及其相关胞外酶活性变化,以了解洛巴伊大口蘑对碳源的降解利用特点。结果表明:洛巴伊大口蘑具有完整的胞外纤维素酶系;淀粉酶在菌丝生长阶段活性较高;羧甲纤维素酶、滤纸纤维素酶和半纤维素酶的活性与纤维素和半纤维素的降解速率呈正相关,但β-葡萄糖苷酶活性与纤维素的降解速率没有相关性,漆酶和过氧化物酶活性与木质素的降解速率没有相关性;洛巴伊大口蘑具有很强的木质素降解能力,属于白腐菌;非木质纤维素主要在菌丝生长阶段被利用,木质纤维素是子实体生长发育阶段的主要碳源;在107 d的栽培过程中,培养基失重52.40%,其中呼吸消耗43.98%,子实体转化率为8.42%。
- Abstract:
- Taking Tricholoma lobayense as material,using bag cultivation method,degradation of the cottonseed shell medium and activity changes of related extracellular enzymes were determined at different stages of development from Tricholoma lobayense cultivated on the cottonseed shell medium in order to reveal the features of degradation of carbon source.The results indicated that T.lobayense had a complete set of extracellular cellulose;the activity of extracellular amylase was comparatively high at mycelial growth stage;there existed the positive correlation between the degradation rates of the cellulose and hemicelulose in the medium and the activities of extracellular carboxymenthel cellulase (CMCase),filter paper cellulase (FPase) and hemicellulase (HCase),but no correlation was found between the degradation rates of the cellulose in the medium and the activity of β-glucosidase,no correlation was found between the degradation rates of the lignin in the medium and the activities of laccase and peroxidase;T.lobayense degraded lignin strongly,so it belongs white-rot fungi;nonlignocellulose was principally used at the stage of mycelial growth,however,lignocellulose were the main carbon source for the fruiting stage of the fungus;at the end of cultivation(107 d),the medium reduction (dry weight) was 52.40%,the respiration consumption was 43.98%,the absolute biological efficiency(biomass of fruitbodies) was 8.42%.
