- 以弄岗唇柱苣苔为试材,采用正交实验设计研究培养基、生长抑制剂、渗透压、光强等对其离体保存的影响,并在此基础上确定常温下最适弄岗唇柱苣苔离体的保存条件,以期建立弄岗唇柱苣苔种质资源离体保存技术体系。结果表明:温度为25℃时,弄岗唇柱苣苔离体保存的最佳方法为1/2MS+蔗糖60 g/L+琼脂4.0 g/L+甘露醇5 g/L+矮壮素(CCC) 1.0 mg/L;最佳光照条件为1 600 lx,12 h/d。试验结果表明,建立的弄岗唇柱苣苔常温离体保存体系,经保存300 d,存活率在50%以上,保存材料生长情况良好,可以用于弄岗唇柱苣苔种质资源离体保存。
- Abstract:
- Taking Chirita longgangensis W.T.Wang in Guihaia germplasm as material,using orthogonal to research the effects of medium,growth inhibitor,phytohormone,osmotic pressure,and the growth recovery were evaluated afer in vitro conservation to establish the optimalin vitroconservation technology system of Chirita longgangensisW.T.Wang in Guihaia germplasm.The results showed that the best conservation condition in 25℃ was 1/2MS basal medium+60 g/L sucrose+4.0 g/L agar powder+5 g/L mannite supplemented+1.0 mg/L CCC,in an illuminated chambr under 12 h photoperiod of 1 600 lx light intensity,more than 50% conservation material survived after 300 days,most of them could growth well on propagation medium.The results of this study could be used for medium-term germplasm preservation in vitroof Chirita longgangensis?W.T.Wang in Guihaia.
