- 以星点木幼嫩茎段为外植体,对其愈伤组织诱导和植株再生进行试验,以期筛选出其在组织培养过程中较合适的激素组合。结果表明:诱导愈伤组织最佳培养基为MS+6-BA 2.0 mg/L+TDZ 0.5 mg/L+2,4-D 1.0 mg/L+蔗糖30 g/L+琼脂6 g/L,其诱导率为88.9%;诱导不定芽分化最佳培养基为MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+蔗糖30 g/L+琼脂6 g/L,其诱导率为59.26%;不定芽增殖最佳培养基为MS+6-BA 1.0 mg/L+NAA 0.8 mg/L+GA30.2 mg/L+蔗糖30 g/L+琼脂6 g/L,其增殖系数为1.91;最佳生根培养基为1/2 MS+IBA 1.0 mg/L+蔗糖15 g/L+琼脂6 g/L,其生根率达81.2%。
- Abstract:
- Taking the stem segments of Dracaena godseffiana as explant,the callus induction and plantlet regeneration were studied,in order to select the suitable hormone combination in the process of tissue culture.The results indicated that:the optimum medium for callus induction was MS+6-BA 2.0 mg/L+TDZ 0.5 mg/L+2,4-D 1.0 mg/L+sucrose 30 g/L+agar 6 g/L,induction rate was 88.9%;the optimum medium for adventitious bud differentiation was MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+sucrose 30 g/L+agar 6 g/L,induction rate was 59.26%;The optimum medium for adventitious bud proliferating was MS+6-BA 1.0 mg/L+NAA 0.8 mg/L+GA3 0.2 mg/L+sucrose 30 g/L+agar 6 g/L,proliferation coefficient was 1.91;The optimum medium for rooting was 1/2MS+IBA 1.0 mg/L+sucrose 15 g/L+agar 6 g/L,rooting rate was 81.2%.
