- 以石韦的干燥叶为试材,采用单因素试验结合响应面法优化石韦总黄酮提取工艺,研究了石韦提取物对ABTS+·、DPPH·、OH·等自由基的抗氧化活性,以期为石韦后续提取工艺及抗氧化能力的开发利用提供参考依据。结果表明:石韦总黄酮的最优提取工艺为料液比1∶40 g·mL-1,提取时间32 min,提取温度 80 ℃。经3次试验验证,总黄酮含量为(120.41±1.25) mg·g-1 (RSD=1.37%,n=3)。体外试验表明,石韦总黄酮对ABTS+·、DPPH·、OH·有一定的清除能力。Box-Behnken响应面法可用于石韦总黄酮的提取工艺优化,提取工艺稳定、可行。该条件下得到的石韦总黄酮提取物有较好的抗氧化活性。
- Abstract:
- Taking dried leaves of Pyrrosia lingua as test materials,the extraction process of total flavonoids from dried leaves of Pyrrosia lingua was optimized by single factor experiment combined with response surface methodology,and the antioxidant activity of the extract against free radicals such as ABTS+·,DPPH· and OH· was studied,in order to provide reference for the followup extraction process and the development and utilization of antioxidant capacity.The results showed that the optimum extraction conditions of total flavonoids were as follows,ratio of material to liquid was 1∶40 g·mL-1,extraction time was 32 minutes,extraction temperature was 80 ℃.After three experiments,the content of total flavonoids was (120.41±1.25) mg·g-1 (RSD=1.37%,n=3).The in vitro test showed that the total flavonoids of Pyrrosia lingua had certain scavenging ability on ABTS+·,DPPH·,OH·.BoxBehnken response surface method could be used to optimize the extraction process of total flavonoids from Pyrrosia lingua,and the extraction process was stable and feasible.Under these conditions,the extract of total flavonoids from Pyrrosia lingua had good antioxidant activity.
