- 以狭叶番泻枝条为试材，采用组织培养的方法，研究不同培养基对狭叶番泻不定芽诱导、增殖、生根诱导以及移栽基质对移栽存活率高低的影响。结果表明：以MS为基本培养基，添加2.0 mg/L TDZ+0.1 mg/L NAA对不定芽的诱导效果较佳，添加2.0 mg/L 6-BA+0.05 mg/L NAA对不定芽的增殖效果最佳，每外植体长出不定芽（12.6±0.64）个；而生根诱导以1/2MS+2.0 mg/L IBA+1%(w/v)活性碳的效果最好，生根诱导率达58.5%。以黄土和泥炭土为基质，移栽成活率为73.7%，移栽苗与种子苗在田间生长表现无明显差异。该研究为狭叶番泻植株扩繁提供了理论基础。
- Abstract:
- Adventitious bud differentiation，bud seedling multiplication，root induction from mature brunch of Cassia angustifolia，and transplanting substrate of tissue culture-raised plantlets transplantation were studied by tissue culture method.The results showed that the efficient adventitious bud differentiation was cultured on MS media supplemented with 2.0 mg/L TDZ+0.1 mg/L NAA.The highest number of adventitious buds (12.6±0.64) per explant were observed in MS media supplemented with 2.0 mg/L 6-BA+0.05 mg/L NAA.The highest rooting percentage (58.5%) and survival were achieved using 1/2MS media with 2.0 mg/L IBA and 1% (w/v) active carbon.The 73.7% plantlets survived acclimatization in the greenhouse，and producing healthy plants on the field.The current investigation showed efficient in vitro regeneration capabilities of Cassia angustifolia from brunch explants.
