- 以不同品种的朱顶红(Hippeastrum rutilum)发病组织为试材,采用组织分离法对病原菌进行分离,并对分离出的病原菌进行了形态学观察和ITS序列分析鉴定,并探究了3种不同浓度(0.01、0.02、0.04 g·mL-1)的5种农药(百菌清、多菌灵、代森锰锌、甲基硫菌灵、菌核净)对分离出的病原菌的体外抑制作用,以期为朱顶红根腐病病原菌的分离鉴定及防治提供参考依据。结果表明:从朱顶红发病样品中共分离出了25个单菌株,根据菌落性状分为3类菌,ITS序列分析发现25个单菌株实际上只有3种基因序列,通过BLAST初步鉴定均属于镰刀菌属(Fusarium)。此外,5种农药对3种朱顶红根腐病病原菌的体外抑制作用不同,其中有7个处理组具有不同程度的显著性抑制作用。
- Abstract:
- Taking different varieties of Hippeasterum rutilum diseased tissues as the test materials,the pathogenic bacteria were isolated by tissue isolation method,and the isolated pathogenic bacteria were observed morphologically and identified by ITS sequence analysis.The in vitro inhibitory effects of five pesticides (hlorothalonil,carbendazim,mancozed,thiophanate-methyl,and dimetachlone) at three different concentrations (0.01,0.02,0.04 g·mL-1) on the isolated pathogenic bacteria were explored,in order to provide reference for the isolation,identification,and prevention of root rot pathogens in Hippeasterum rutilum.The results showed that a total of 25 single bacterial strains were isolated from the samples of the Hippeastrum rutilum diseased plant,which were classified into three types of bacteria based on their colony characteristics.ITS sequence analysis revealed that the 25 single bacterial strains actually only had three gene sequences,and were preliminarily identified by BLAST as belonging to the genus Fusarium.In addition,the in vitro inhibitory effects of five pesticides on three types strains of root rot disease from Hippeastrum rutilum were different,with seven treatments showing significant inhibitory effects to varying degrees.
