双城洋葱软腐病原的分离鉴定

- 以双城洋葱种植地贮存期感病洋葱为试材,采用组织分离、柯赫法则、形态、生理生化和分子鉴定方法,对病原菌进行了分离鉴定,并研究了病原菌致病谱,以期为洋葱种植与贮存中软腐病的防治提供参考依据。结果表明:洋葱软腐病原是一株细菌YC1,寄主较广,还可引起芹菜、马铃薯、白菜、胡萝卜和青椒腐烂。该菌株为G-杆菌,可利用葡萄糖、果糖、海藻糖、蔗糖、山梨醇、甘露醇、柠檬酸盐和丙二酸盐,不能分解淀粉和纤维素,氧化酶、吲哚、H2S和V.P反应阴性,M.R、接触酶、石蕊牛奶、硝酸盐还原和明胶液化阳性。16S rDNA、gyrB和rpoD基因序列系统进化分析显示与Burkholderia gladioli亲缘关系最近。综合形态、生理生化特性以及16S rDNA、gyrB和rpoD基因序列分析鉴定该病原菌为B.gladioli.

- The diseased onions in Shuangcheng onion plantation were used as test materials,the pathogen causing onion soft rot was isolated by tissue isolation and Coch rule,and identified based on morphological,physiological and biochemical characteristics as well as 16S rDNA,gyrB and rpoD sequence analysis,in order to provide reference for the prevention and control of onion soft rot disease.The results showed that a pathogenic bacterium YC1 was isolated from the infected onion tissue.The strain exhibited the strong and widEspectrum pathogenicity.It could also infect cabbage,carrot,pepper,celery and potato tissue.The utilized glucose,fructose,sucrose,malonate,trehalose,sorbitol,mannitoland citrate,did not hydrolyze starch andcellulose.oxidase,H2S,indole and V.P reactions were negative.M.R,contact enzyme,litmus milk,nitrate reduction and gelatin liquefaction were positive.Phylogenetic analysis based on 16S rDNA,gyrB and rpoD sequence showed that the strain had high homology with B.gladioli.The strain YC1 was identified as B.gladioli according to morphological,physiological and biochemical characteristics and 16S rDNA,gyrB and rpoD sequence analysis.
