- 以非洲菊根腐病发病植株为试材,采用常规组织分离法对引起非洲菊根腐病病原菌进行分离纯化,研究了分离物对非洲菊的致病性,并通过形态学结合 rDNA-ITS和TEF-1α序列的分子系统学方法对病原菌进行鉴定,以期明确非洲菊根腐病的病原菌类型。结果表明:从典型病根上共分离获得8株菌株,分别为FZJf 1-1、FZJf 1-2、FZJf 1-3、FZJf 2-1、FZJf 2-2、FZJf 2-3、FZJf 3-1和FZJf 3-2;8株菌株均能导致非洲菊幼苗出现典型的叶片枯萎和根腐症状,其中菌株 FZJf 1-1和FZJf 1-3致病力最强,接种第3周植株开始发病,其次是菌株FZJf 2-1,接种第5周植株开始发病,其余菌株致病性较弱;根据形态学特征和分子系统学特征鉴定出6 株为尖孢镰刀菌(Fusarium oxysporum),分别是FZJf 1-1、FZJf 1-2、FZJf 1-3、FZJf 2-1、FZJf 2-3和FZJf 3-2,1 株为茄类镰刀菌(F.solani)FZJf 2-2,1 株为层出镰刀菌(F.proliferatum)FZJf 3-1,尖孢镰刀菌(F.oxysporum)为优势病原菌,茄类镰刀菌(F.solani)和层出镰刀菌(F.proliferatum)为弱势病原菌。
- Abstract:
- The disease plants of gerbera root rot were used as the experimental materials,pathogen species were isolated and purified from the gerbera root rot by the tissue isolation method,then the pathogenicity of the isolates to gerbera plants were studied,finally the pathogen species were identified by using morphological and molecular systematics method based on rDNA-ITS and TEF-1α sequences,in order to make clear the fungal pathogen species of thegerbera root rot disease.The results showed that a total of 8 pathogenic strains of FZJf 1-1,FZJf 1-2,FZJf 1-3,FZJf 2-1,FZJf 2-2,FZJf 2-3,FZJf 3-1 and FZJf 3-2 were isolated from severely rot roots;pathogenicity test results showed that all the 8 strains were pathogenic to gerbera seedlings and could cause typical leaf wilt and root rot symptoms,among which strains FZJF 1-1 and FZJF 1-3 had the strongest pathogenicity,the plants developed disease after 3 weeks inoculation,followed by strain FZJF 2-1,the plants developed disease after 5 weeks inoculation,while the other strains were less pathogenicity;6 strains of FZJf 1-1,FZJf 1-2,FZJf 1-3,FZJf 2-1,FZJf 2-3 and FZJf 3-2 were identified as Fusarium oxysporum,one strain of FZJf 2-2 was identified as F.solani and one strain of FZJf 3-1identified as F.proliferatumby morphological and molecular phylogenetic characteristics.F.oxysporum species were the dominant pathogens,and F.solani and F.proliferatum were weak pathogens.
