- 以日本灵芝子实体为试材,在单因素试验的基础上,采用响应面分析方法对微波辅助法提取灵芝多糖工艺进行了优化;以枯草芽孢杆菌、白色念珠菌、金黄色葡萄球菌、大肠杆菌、沙门氏菌为供试菌株,研究灵芝多糖抑菌活性;采用倍比稀释法,确定了最小抑菌浓度。结果表明:微波辅助法最佳工艺为微波功率374 W,提取时间23 min,液料比15∶1 mL·g-1,浸提次数2次,在此条件下灵芝多糖提取率达到了4.62%;灵芝多糖对5种供试菌均表现出抑制作用,其中对金黄色葡萄球菌、大肠杆菌和枯草芽孢杆菌的抑制作用明显,抑菌率最高达到了52.4%;灵芝多糖对金黄色葡糖球菌的最小抑制浓度为6.25 mg·mL-1,对枯草芽孢杆菌和大肠杆菌均无菌落生长且最小抑制浓度为12.50 mg·mL-1;对白色念珠菌的最小抑制浓度为25.00 mg·mL-1,对沙门氏菌的最小抑制浓度为50.00 mg·mL-1。
- Abstract:
- Based on the single factor experiment,the response surface method was used to optimize the extraction process of Ganoderma lucidum polysaccharides by the response surface method.Bacillus subtilis,Monilia albican,Staphylococcus aureus,Escherichia coli and Salmonella were used as test strains to study the antibacterial activity of Ganoderma lucidum polysaccharides.The minimum inhibitory concentration was determined by double dilution method.The results showed that microwave power was the best process of microwave power 374 W,extraction time 23 minutes,liquid material 15∶1 mL·g-1,extraction times 2 times,under this condition,the extraction rate of Ganoderma lucidum polysaccharides reached 4.62%.Ganoderma lucidum polysaccharides showed inhibitory effects on five kinds of tested bacteria,of which Staphylococcus aureus,Escherichia coli and Bacillus subtilis significantly inhibited the maximum inhibitory rate reached 52.4%.The minimum inhibitory concentration of Ganoderma lucidum polysaccharide against Staphylococcus aureus was 6.25 mg·mL-1,and the minimum inhibitory concentration against Bacillus subtilis and Escherichia coli was 12.5 mg·mL-1.The minimum inhibitory concentration against Monilia albican was 25.00 mg·mL-1 and the minimum inhibitory concentration against Salmonella was 50.00 mg·mL-1.
