- 以北京地区采集的野生羊肚菌为试材,对其子囊果进行组织分离,采用CTAB法提取羊肚菌DNA,琼脂糖电泳检测,ITS序列测定和构建分子系统发育树,通过分离鉴定比较北京山区野生羊肚菌,以期为羊肚菌的人工栽培和菌种资源利用提供依据。结果表明:分离到羊肚菌菌株7份,各菌株亲缘关系很近,菌丝形态相似,但颜色深浅、菌核多少、菌丝生长速度明显不同。系统发育分析表明,所采集的北京野生羊肚菌均属于黄色羊肚菌类群。
- Abstract:
- The wild Morchella collected in Beijing was used as test material,by tissue separation method,CTAB method was used to extract Morchella DNA,agarose electrophoresis,ITS sequencing and molecular phylogenetic tree were conducted.Wild Morchella in Beijing mountainous area were isolated and identified,in order to provide basis for artificial cultivation of Morchella esculenta and utilization of microbial resources.The results showed that 7 isolates of Morchella were successfully isolated.The phylogenetic relationship of the isolates was very close.The mycelial morphology were similar,but the color depth,number of sclerotia,and mycelial growth rate were significantly different.Phylogenetic analysis showed that the wild morch collected in Beijing belonged to the yellow morel group.
