- 以保山野生红蜡蘑为试材,采用微波辅助法提取了红蜡蘑多糖,在单因素试验的基础上,采用Box-Behnken响应面法对多糖提取的显著因素:微波时间、微波功率、液料比进行了优化;用对1,1-二苯基-2-三硝基苯肼自由基(DPPH·)和羟自由基(·OH)的清除率测定红蜡蘑多糖的抗氧化活性。结果表明:多糖提取的最佳工艺为微波时间90 s,微波功率350 W,液料比50∶1 mL·g-1,水浴温度80 ℃,水浴时间2 h,多糖平均得率为6.75%,与预测值(6.79%)相比,相对误差较小,为0.59%;红蜡蘑多糖对DPPH·和·OH具有明显的清除能力,红蜡蘑多糖是一种具有抗氧化能力的食用真菌多糖,值得进一步重视和研究。
- Abstract:
- The wild Laccaria laccata in Baoshan was used as the material,the polysaccharides was extracted by microwave assisted method,based on the single factor experiment,the response surface method of Box-Behnken significant factors of microwave time,microwave power and liquid-solid ratio.The antioxidation of polysaccharides was determined by the scavenging effect of DPPH· and ·OH.The results showed that the optimum conditions of Laccaria laccata polysaccharides were:microwave time 90 seconds,microwave power 350 W,liquid-solid ratio 50∶1 mL·g-1,water bath temperature 80 ℃,water bath time 2 hours,and the polysaccharide yield was 6.75%.Compared with predicted value (6.79%),it had a smaller relative error at 0.59%.Furthermore,Laccaria laccata polysaccharides had remarkable scavenging activity to DPPH· and ·OH,which indicated that was a strong natural antioxidation and further study was worth doing.
