- 以白刺花无菌苗叶片为试材,利用双因素正交实验设计,研究了不同生长调节剂对愈伤组织诱导效果的影响,以期筛选最佳的诱导培养基,为白刺花组培研究提供参考基础。结果表明:在MS培养基上,2,4-D与6-BA组合可诱导出较疏松型的愈伤组织,愈伤组织表面有瘤状突起且部分愈伤上有绿色芽点;2,4-D与KT组合也可诱导形成愈伤组织,但愈伤组织的生长及疏松度不及2,4-D与6-BA组合效果明显;NAA与6-BA,NAA与KT组合早期培养时在叶片边缘看到部分愈伤的生长,培养45 d后愈伤开始褐化;在基本培养基研究筛选上,MB和B5均能诱导愈伤组织,愈伤组织颜色稍显淡绿。2,4-D有利于白刺花愈伤组织的诱导,最佳愈伤组织诱导培养基为MS+2,4-D 1.0 mg?L-1+6-BA 1.0 mg?L-1+蔗糖4%+琼脂0.7%,出愈率75.58%,愈伤组织出愈早,疏松,生长旺盛。
- Abstract:
- To establish excellent callus induction culture system,the no vaccine leaves of Sophora davidii were used as materials to research effect of different growth regulators on callus induction by double factors orthogonal design of experiment.The aim was to screen the best callus induced medium in order to provide a theoretical basis for tissue culture of Sophora davidii. The results showed that 2,4-D and 6-BA combination could induce relatively loose callus on the MS culture medium.Callus surface had protrusions and green bud points on the part of the callus.2,4-D combined with KT could also induce the formation of callus,but the growth and loose degree of callus was less than 2,4-D and 6-BA combination.NAA and 6-BA combination,NAA and KT combination could induce callus formation early in the leaf edge,callus brown gradually after culture 45 days.MB and B5 could induce callus,callus color was slightly green on the basic medium.2,4-D was advantageous to callus induction of Sophora davidii. The best callus induction medium was MS+2,4-D 1.0 mg?L-1+6-BA 1.0 mg?L-1+4% sucrose+0.7% agar,the induction rate was 75.58%,callus grew earlier,osteoporosis,vigorous growth in the whole experiment.
