- 以2种从美国引种的藤蔓月季Climbing rosa ‘Alchymist’ 和 ‘Handel’ 为试材,采用正交设计进行扦插试验,比较不同扦插基质和外源激素处理组合的生根差异,结合发根过程过氧化物酶(POD)、吲哚乙酸氧化酶(IAAO)和多酚氧化酶(PPO)活性变化,分析它们的变化与生根率的关联性。结果表明:‘Alchymist’扦插生根的较佳配方为200 mg?L-1 ABT,黄土∶河沙=1∶1,生根率达到86 67%;‘Handel’在200 mg?L-1 IBA,黄土∶腐殖土=1∶1的配方中生根率最高,为96 67%。‘Alchymist’和‘Handel’的POD活性基本呈现上升-下降-上升-下降趋势。2个品种的POD活性在愈伤组织形成时持续上升,POD活性的上升率与生根率成正比,‘Alchymist’生根率最高的处理中POD活性上升率最大,上升率达363 96%。2个品种的PPO活性变化呈现上升-下降趋势,不定根产生过程中,PPO活性一直上升,且‘Alchymist’和‘Handel’的PPO活性上升率与生根率成正比,‘Handel’生根率最高的处理PPO活性上升率最大,达661 423%。2个品种的IAAO活性变化大体呈下降-上升-下降趋势,不定根产生过程中,2个品种IAAO活性与生根率呈负相关,‘Alchymist’生根率最好的处理IAAO活性下降率最大,达45 53%。‘Alchymist’和‘Handel’生根率与POD活性和PPO活性呈正相关,与IAAO活性呈负相关。‘Alchymist’和‘Handel’的生根率与3种酶的相关系数排列顺序为0 664(PPO)>0 606(POD)>0 463(IAAO)。
- Abstract:
- In order to compare the rooting difference under the different treatments,an orthogonal design was carried out to analyze the rooting effects of combinations by the mediums type,hormone kinds and different concentrations.Meanwhile,the change of physiological indexes,which involved three enzymes of POD,IAAO,PPO,and their correlations with the rooting rate were detected at the rooting process.The results indicated that the rooting rates of ‘Alchymist’ could reach to 86 67% under this formula(200 mg?L-1 ABT,yellow soil∶sand=1∶1).However,‘Handel’ had the highest rooting rate of 96 67% with this combination(200 mg?L-1 IBA,yellow soil∶humus soil=1∶1).The tendency of POD activity was kept up-down-up-down in testing period.The POD activity maintained increasingly at the callus formation stage and had positively affected on the rooting rate.‘Alchymist’ obtained the highest rooting rate when its activity dramatically increased to 363 96% to get the maximum value.Moreover,the PPO activity had a up-down tread in the rooting stage.The PPO activity had constantly increased in the process of adventitious roots and had positively affected on the rooting rate.The activity was the highest in the best treatment of rooting rate,which reached 661 432% for ‘Handel’.However,the tendency of IAAO activity was down-up-down for both.The declining rate of IAAO activity was directly proportional to the rooting effect.Additionally,activity of POD and PPO had positively correlated with the rooting effect,but it had a negative correlation with the activity of IAAO.The correlation coefficients between the rooting rate and three physiological indexes were listed as followed,0 664(PPO)>0 606(POD)>0 463(IAAO).
