- 以芦笋高产品种“井岗701”的无菌茎尖为试材,采用正交实验设计方法,研究嘧啶醇、KT、2,4-D、脱落酸、6-BA和NAA等不同植物激素浓度组合对愈伤组织、胚状体以及幼苗诱导的影响。结果表明:MS+2,4-D 2.0 mg/L+KT 1.0 mg/L诱导愈伤组织效果最好,出愈率达到80%左右。MS+0.70 mg/L嘧啶醇+0.10 mg/L NAA+0.50 mg/L KT诱导胚状体效果最好,诱导率高达95%。MS+NAA 0.5 mg/L+6-BA 1.0 mg/L诱导胚状体成苗率达到60%左右。室外移栽成活率高达90%,初步建立了芦笋由胚状体到幼苗的一步成苗体系。
- Abstract:
- The sterile shoot tips of high-yielding varieties of ‘Jinggang 701’ Asparagus officinals were used as experimental materials,using orthogonal design,the effect of different concentrations combinations of pyrimidinol,KT,2,4-D ,abscisic acid,6-BA and NAA on the inductions for callus,somatic embryoids and seedlings were studied.The results showed that the optimum inducing medium for callus was MS+2,4-D 2 mg/L+KT 1.0 mg/L,the inducing rate reached to 80%;the optimum inducing medium for embryos was MS+0.70 mg/L ancymidol+0.10 mg/L NAA+0.50 mg/L KT,the inducing rate reached to 95%;the optimum inducing medium for plantlet was MS+NAA 0.5 mg/L+6-BA 1.0 mg/L ,the inducing rate reached to 60%;the survival rate of outdoor transplant could be about 90%.The system of one-step inducing into plantlet for asparagus was initially established.
