- 以豇豆为试材,用新型植物基因组DNA提取试剂盒提取豇豆DNA,选取Mg2+、dNTPs、TaqDNA聚合酶、引物及模板DNA为主因素进行正交优化试验,从3个不同循环次数中筛选出最佳效果循环次数。结果表明:优化的反应体系为25μL的反应体系中Mg2+1.0mmol/L、dNTP0.2μmol/L、TaqDNA聚合酶1.5U、引物0.3μmol/L、模板DNA45ng,循环次数为40次。

- GenomicDNAofasparagusbeanwasextractedbythemethordofDNAsecurePlantkit.FivefactorssuchasconcentrationofMg2+,dNTPs,TaqDNA,primer,andtemplateDNAwhichwereprimaryfactorfortheorthogonaldesignwasselected,whilethebestcyclesofPCRreactionwasalsoscreenedfromthreedifferenttrements.TheresultsshowedthattheoptimizationforISSR-PCRreactionsystemonasparagusbeanwasasfollows:25μLreactionsystemcontaining1.0mmol/LMg2+,0.2μmol/LdNTP,1.5UTaqDNApolymerase,0.3μmol/Lprimerand45ngDNAtemplet,thewholePCRreactionranfor40cycles.
