Abstract:
Abstract:InordertoestablisharapidpropagationsystemofD.primulinum,immatureseedsofDendrobiumprimulinumLindl.wereusedasexplantsfortissueculture,andmediumsforasepticseedgermination,subcultureofprotocom,rootingandgrowthofstrongplantletswerescreened.TheresultsshowedthatthemediumMS+6-BA10mg?L-1+NAA01mg?L-1+sugar20g?L-1wasoptimumforseedgermination.TheoptimumsubculturemediumthatforproliferationofprotocormandshootinductionwasMS+6-BA10mg?L-1+NAA01mg?L-1+mashedbanana100g?L-1+sugar20g?L-1,thepropagationcoefficientwas82aftersubcultured45days.TheoptimumrootingmediumwasMS+6-BA10mg?L-1+KT05mg?L-1+NAA02mg?L-1+mashedbanana100g?L-1+sugar30g?L-1+activatedcarbon10g?L-1,therootingratereached100%andtherootshadhigherqualityandmorequantityaftercultured90days.ThebesttimeforthetransplantationofrootregenerationplantswasMarchtoAprilinGuilinarea,therootedplantsgrewbetterinmediaⅠ(pinebark∶sphagna∶perlite∶vermiculite=10∶2∶1∶1)thanthatinmediaⅡ(pinebark∶perlite∶vermiculite=10∶1∶1),andthesurvivalratewas923%.
