Abstract:
Fusarium species were obtained from wilted bitter gourd samples collected from six bitter gourd fields of Zhengzhou,Zhoukou and Xinyang in Henan Province using the method of tissue isolation.In order to clarify the classification status of the Fusarium species in Henan Province and provide a reference for Fusarium wilt prevention.Isolates were identified via the culture features,the morphological characteristics of colony and spore,molecular phylogenetic analysis and pathogenicity.The results showed that all isolates were identified as Fusarium oxysporum f.sp.momodicae Sun & Huang.These isolates could infect the seedlings of bitter gourd,and luffa,bottle gourd weakly,but not pumpkin,muskmelon,watermelon and cucumber.Phylogenetic tree analysis based on multiple gene sequences of ITS,[STBX]EF-1α[STBZ] and β-tubulin showed that the strains of Fusarium oxysporum from Zhengzhou and Xinyang had closer genetic relationship with the above specialization.Except HeN2 from Zhengzhou,all isolates from Zhengzhou and Xinyang were clustered to the same subgroup with Fusarium oxysporum f.sp.lagenariae,Fusarium oxysporum f.sp.niveum,and Fusarium oxysporum f.sp.melonis.The isolates from Zhoukou could be significantly divided into 2-3 subgroups,of which,HeN25,HeN27,HeN28,HeN30 and HeN32 belonged to the same subgroup and had closer genetic relationship with Fusarium oxysporum f.sp.cucumerinum.HeN18,HeN26,HeN29,HeN31,HeN33 and HeN34 had closer genetic relationship with Fusarium oxysporum f.sp.cubense
