Abstract:
Lilium oriental ‘Sorbonne’ diploid cultivars were used as material to explored the method and possibility of stained with colchicine package method to induce 2n gamete.In order to get the best induced concentration and time,taking leaf from the same plant as control,2n pollen were authenticated by flow cytomety instrument.The results showed that the highest mutation rate of flower buds was 68%,the best concentration of colchicines was 0.4%,and the handling time was 24 hours in the meiosis stage I of the pollen mother cell through package method.The abnormal phenomena like days and triads at the tetrad stage and cytoplasm separation but the chromosome non separation,etc.were observed,which was identificated by flow cytometry.The results obtained that the 2n pollen was diploid as the monoecious plants for the control.The results implied that 2n pollen induced artificially can be a powerful method to creat the sexual polyploids of Lilium oriental.
