Abstract:
Taking7varietiesofZantedeschiahybridaasmaterials,usingcolorcallalily‘Prafait’DNAastemplateDNA,theconcentrationofMg2+,Primer,dNTPsandtemplateDNA,whichgreatlyinfluencetheRAPD-PCR,wereoptimizedbyUniformDesignmethodU12(34).TheresultsshowedthatoptimumRAPDreactionsystemofZantedeschiahybridawasevery20μLreactionsystemcontaining1×PCRBuffer,1.5mmol/LMgCl2,0.55μmol/Lprimer,0.125mmol/LdNTPs,25ngDNAtemplateand1.0UTaqDNApolymerase.
