Abstract:
ChosenyoungleavesofasepsisseedlingofLactucasativaL.astestmaterial,theeffectofmaterialresources,plasmolysistime,seedlingage,enzymecombination,enzymolysistimeandostimumpressureonprotoplastisolationwerestudied.Theresultsshowedthatwhenyoungleaveswereplasmolysedfor2hinCPWsolutioncontaining13%mannitol,theyieldofprotoplastanditssurvivalratewerehigherthanthoseofotherplasmolysistime.20doftheyoungleaveswereisolatedprotoplastshighquality,protoplasmtakestheshapeofcircular,manyinclusion,strongvitality;Theenzymeconcentrationwas1.0%cellulase+0.2%macerozyme,isolateezyme.Alargeamountofhigh-activeprotoplastcouldbeobtainedbyenzymolysisfor12hat25℃underdarkconditions.Theminstraof9%weresuitableforprotoplastdissociation.
