Abstract:
The semi-hardwood stem segments of southern high-bush blueberry ‘Biloxi’ were used as explants,and the optimum conditions for tissue culture were optimized by orthogonal test.The results showed that the best disinfection method for explants was to disinfect them with 75% ethanol for 45 seconds,then 10% NaClO for 10 minutes,and then put stems into WPM+ZT 1.00 mg·L-1 medium;the optimum medium for propagation culture was WPM+6-BA 1.00 mg·L-1+ZT 2.00 mg·L-1+IBA 0.25 mg·L-1;the optimum medium for strong seedling culture was WPM+ZT 0.50 mg·L-1+IBA 0.50 mg·L-1;and the optimum medium for rooting was WPM+vermiculite∶coconut coir (volume ratio 1∶1)+activated carbon 3.00 g·L-1+IBA 0.500 mg·L-1.
