Abstract:
WRKYdomain proteins play important roles in plant stress responses.The salttreated tomato was used as material,the sequence and expression pattern of tomato SlWRKY44 gene were explored by using RTPCR and realtime PCR technology,in order to provide a reference for studying the function of this gene in salt stress response.The results showed that the full length SlWRKY44 CDS was 984 bp.This CDS sequence encoded a polypepide of 327 amino acid residues with a predicted molecular mass of 36.72 kDa and a basic isoelectric point of 5.79,and a highly conserved WRKY domain in the encoded putative protein.Protein subcellular localization predicted indicated that SlWRKY44 was localized in the nucleus.Phylogenetic analysis indicated that SlWRKY44 clustered with AtWRKY46,AtWRKY53 and AtWRKY54.Tissuespecific expression profiles showed that SlWRKY44 was nearly expressed in all detected tissues,and its expression level was relatively high at root and low at stem.Quantitative RTPCR analysis showed that SlWRKY44 was induced by salt stress.The expression patterns of SlWRKY44 under salt stress suggested that this gene might be involved in the regulations of tomato to salt stress.
