- Abstract:
- The full buds and shoot segments of healthy plants were used for in vitro culture,a complete and practical micropropagation protocol for rapid vegetative propagation of the rootstock ‘St.Julien A’ was developed.The results showed that using fully developed buds as explants,MS+BA 1.0 mg·L-1+IBA 0.1 mg·L-1 as the initial medium,WPM+BA 1.0 mg·L-1+IBA 0.1 mg·L-1 as proliferation medium and 1/2MS+IBA 1.0 mg·L-1 as rooting medium,the proliferation coefficient and the rooting percentage reached 8.5 and 96%,respectively.Transplanted in the substrate of vermiculite,the survival percentage of transplanting plantlets reached 91.6%.
