- 以河北沽源、北京房山辛庄2个地区生产的‘Tresor’百合种球为试材,采用鳞片扦插的方法,研究了不同地理产区和不同储存周期采收的百合种球的激素含量(CTK、GA3、ABA、IAA)、营养成分(包括蛋白质、可溶性蛋白质、总多糖、葡萄糖、蔗糖、淀粉)以及酶活性(SS-〖KG-*4〗Ⅰ、SS-〖KG-*4〗Ⅱ、α-淀粉酶、β-淀粉酶)的各项生理指标对‘Tresor’百合鳞片扦插的影响,以期为进一步提升百合种球繁育技术提供参考依据。结果表明:河北沽源和北京房山生产的百合种球因气候差异明显,导致采收的百合种球在营养成分含量、淀粉酶活性以及内源激素水平存在一定差异,但鳞片扦插后微鳞茎发生的起始时间以及所有鳞片完成微鳞茎发生需要的时间长短没有明显差异。采收后经过一定低温储存的百合鳞片的营养成分含量、淀粉酶活性以及内源激素水平会随着低温储存的时间延长而发生改变,不同储存周期的微鳞茎发生起始时间没有明显差异,均在12~14 d,但随着储存时间的增加,微鳞茎发生周期从最长36 d(种球储存3周)逐渐缩短至22 d(储存12周,种球休眠解除),随后又延长至26 d(储存16周)。以上结果表明,百合微鳞茎的发生与采后鳞片初始营养成分含量、酶活性以及内源激素水平等生理指标无相关性,而低温储存周期的长短直接影响鳞片扦插后微鳞茎的发生速率和周期,存储12周的百合种球扦插效率最高。
- Abstract:
- Taking lily bulbs of ‘Tresor’ produced in two regions,Gu Yuan in Hebei and Xinzhuang in Fangshan,Beijing,as experimental materials,the effects of various physiological indicators,including hormone contents (CTK,GA3,ABA,IAA),nutritional components (such as protein,soluble protein,total polysaccharides,glucose,sucrose,starch),and enzyme activities (SS-Ⅰ,SS-Ⅱ,α-amylase,β-amylase) on the scale cutting propagation of ‘Tresor’ lilies under different geographical origins and storage periods were studied by using scale cutting method,in order to provide reference for further improving lily bulb breeding technology.The results showed that the significant climatic differences between Guyuan and Fangshan resulted in variations in the nutrient content,starch enzyme activity,and endogenous hormone levels of the harvested lily bulbs.However,the initial time for microbulb formation and the duration required for all scales to complete microbulb formation showed no significant differences.Additionally,the nutrient content,enzyme activity,and endogenous hormone levels of lily scales changed with extended cold storage.The initiation time of microbulb formation did not vary significantly across storage periods,remaining at approximately 12-14 days.However,the microbulb formation cycle gradually shortened with prolonged storage,from a maximum of 36 days (bulbs stored for 3 weeks) to 22 days (12 weeks of storage,dormancy broken),before increasing to 26 days (16 weeks of storage).These findings suggest that the occurrence of lily microbulbs is not correlated with the initial nutrient content,enzyme activity,or endogenous hormone levels of the scales post-harvest.Instead,the length of the cold storage period directly impacts the rate and duration of microbulb formation after scale cutting.Bulbs stored for 12 weeks exhibited the highest propagation efficiency.
