- 以长叶兰(Cymbidium erythraeum Lindl.)×大雪兰(Cymbidium mastersii Griff.ex Lindl.)杂交种子为试材,采用体视显微镜测定种子形态与有胚率,并比较在5种基础培养基(MS、1/2MS、1/4MS、KC、花宝1号)上的无菌萌发及早期原球茎形成情况,以筛选出最佳培养基,以原球茎为外植体,设置3组处理,暗培养24 h(A组)、光照12 h暗培养12 h(B组)及光照12 h+6-BA(1.0 mg·L-1)+NAA(0.5 mg·L-1)(C组),培养120 d后测定原球茎增殖系数与分化率,以期为长叶兰与大雪兰杂交种的种质资源的快速繁殖及育种提供参考依据。结果表明:大雪兰(♂)×长叶兰(♀)种子显著长于大雪兰(♀)×长叶兰(♂),但胚的大小基本一致,有胚率差异不明显,分别为93.65%与97.65%。KC培养基均利于正反交种子萌发及原球茎形成,C组增殖系数及分化率最高,正反交的增殖系数分别为4.50±0.44、18.00±1.09;分化率分别为0.41%±0.05%、0.80%±0.06%。
- Abstract:
- Taking hybrid seeds of Cymbidium erythraeum Lindl.(♀)×Cymbidium mastersii Griff.ex Lindl.(♂) as test material,seed morphology and embryo rate were determined using a stereomicroscope.The study compared sterile germination and early primary bulbil formation on five basic media (MS,1/2MS,1/4MS,KC,and Huabao No.1) to screen for the optimal medium.Using the primary bulbils as explants,three treatment groups were set up,dark cultivation for 24 hours (Group A),12 hours light+12 hours dark (Group B),and 12 hours light+6-BA (1.0 mg·L-1)+NAA (0.5 mg·L-1) (Group C).After 120 days of cultivation,the primary bulbil proliferation coefficient and differentiation rate were measured,in order to provide reference for the rapid propagation and breeding of hybrid germplasm resources of Cymbidium erythraeum and Cymbidium mastersii.The results showed that the hybrid of Cymbidium mastersii (♂)×Cymbidium erythraeum (♀) produced significantly larger seeds than the backcross of Cymbidium mastersii (♀)×Cymbidium erythraeum (♂),though the embryo size was similar,with no significant difference in embryo rate (93.65% and 97.65%,respectively).The KC medium promoted both seed germination and primary bulbil formation for both crosses,with Group C showing the highest proliferation coefficient and differentiation rate.The proliferation coefficients for the two crosses were 4.50±0.44 and 18.00±1.09,respectively,and the differentiation rates were 0.41%±0.05% and 0.80%±0.06%.
