- 以77份葡萄种质为试材,采用SRAP分子标记,从DNA分子水平进行分析,使用非加权组平均法对葡萄种质进行聚类分析,研究了77份葡萄种质的遗传多样性;并根据蒙特卡洛模拟算法进行群体结构分析,以期为推动葡萄分子辅助育种提供参考依据。结果表明:运用SRAP分子标记技术,77份葡萄种质共检测出67个等位基因,每对引物平均扩增出等位基因6.70个,其中有效等位基因5.71个,遗传多样性分析所得遗传多样性指数为0.388 2,香农指数平均为0.563 7,77份葡萄品种试材通过聚类分析分为5类,群体分析为3类。综上所述,77份葡萄种质展现出丰富的遗传多样性。
- Abstract:
- Taking 77 grape germplasms as the test materials,the genetic diversity of the 77 grape germplasms was analyzed at the DNA molecular level using the SRAP molecular marker.The unweighted pair group method with arithmetic mean (UPGMA) was used for cluster analysis of the grape germplasms.The genetic diversity was also analyzed using the Monte Carlo simulation algorithm for population structure analysis,in order to provide reference for promoting molecular assisted breeding of grapes.The results showed that with the SRAP molecular marker,a total of 67 alleles were detected in the 77 grape germplasms.Each primer pair amplified an average of 6.70 alleles,of which 5.71 were effective alleles.The genetic diversity index obtained from the genetic diversity analysis was 0.388 2,and the average Shannon index was 0.563 7.The 77 grape varieties were divided into five categories through cluster analysis,and the population analysis showed three categories.In conclusion,the 77 grape germplasms exhibited rich genetic diversity.
