- 以4种状态的仙客来种子(干种子、吸涨种子、即将萌动种子、萌动种子)为试材,进行不同浓度(0%、0.3%、0.6%、0.9%)、不同时间(4、8、12、16 h)的甲基磺酸乙酯(EMS)浸种处理,分析EMS浸种对仙客来种子萌发率、成苗率、相对成苗率及对胚根和幼苗形态的影响,筛选出适宜的EMS诱变条件,以期为加快仙客来新品种选育进程提供参考依据。结果表明:EMS处理显著抑制仙客来种子的萌发,且随处理浓度升高和处理时间延长,抑制作用越显著;经0.6%EMS处理吸涨种子16 h后,种子未见萌发,全部致死;随着EMS浓度变高、处理时间变长,种子发芽的起始时间推迟、发芽持续时间变长、胚根生长受到的抑制越显著;EMS处理后,仙客来幼苗表现出矮小、生长缓慢、叶片黄化、畸形、卷曲等特殊变异性状。综上,仙客来种子诱变的最佳条件为0.9%EMS浸泡萌动状态仙客来种子4 h。

- Taking four types of Cyclamen persicum seeds in different states (dry seeds,imbibed seeds,seeds on the verge of germination,germinated seeds) as experimental materials,the seeds were treated by soaking in ethyl methanesulfonate (EMS) at different concentrations (0%,0.3%,0.6%,0.9%) for different durations (4,8,12,16 hours).The effects of EMS seed soaking on the germination rate,seedling formation rate,relative seedling formation rate of Cyclamen persicum seeds,as well as on the morphology of radicles and seedlings were analyzed to screen out suitable EMS mutagenesis conditions,in order to provide references for accelerating the breeding process of new Cyclamen persicum varieties.The results showed that the EMS treatment significantly inhibited the germination of Cyclamen persicum seeds,and with the increase of the treatment concentration and the prolongation of the treatment time,the inhibitory effect became more significant.When the seeds were in the imbibed state,all of them were killed after being treated with 0.6%EMS for 16 hours.As the EMS concentration increased and the treatment time became longer,the initial germination time of the seeds was postponed,the germination duration became longer,and the inhibition of radicle growth became more significant.Through morphological observation,it was found that Cyclamen persicum seedlings exhibited special variant traits such as dwarfism,slow growth,leaf chlorosis,deformity,curling,etc.The optimal condition for EMS mutagenesis of Cyclamen persicum was the germinated state seeds soaking in 0.9%EMS for 4 hours.
