- 以辽宁阜蒙县具有根腐病症状的苍术病株为试材,采用组织分离法对病原菌进行分离;根据科赫氏法则对分离菌株进行致病性测定,通过形态学和分子生物学鉴定病原菌种类,并研究其生物学特性;采用菌丝生长速率法测定了病原菌对11种杀菌剂的敏感性,以期为苍术根腐病病害有效防治提供参考依据。结果表明:苍术根腐病主要致病菌为尖孢镰孢菌Fusarium oxysporum,其最适培养基为Richard和察氏培养基,最适氮源为硝酸钾和硝酸钠,最适碳源为蔗糖、甘露醇和麦芽糖。室内药剂试验结果显示,已唑·四霉素和福·霜·敌磺钠对苍术根腐病病原菌抑制效果最好,EC50值<1 mg·L-1,其次是甲霜·噁霉灵。
- Abstract:
- Taking Atractylodes lancea plants with root rot from Fumeng country,Liaoning Province as experimental materials,the pathogen was isolated and purified by using tissue isolation method.Based on Koch′s Postulates,the pathogenicity of the isolated strains was determined.The pathogen species were identified through morphological and molecular biological methods,and their biological characteristics were studied.The sensitivity of the pathogen to 11 fungicides was determined using the mycelial growth rate method,in order to provide reference for the effective prevention and control of root rot disease in Atractylodes lancea.The results showed that the pathogen causing Atractylodes lancea root rot was Fusarium oxysporum.The biological characteristics of the pathogen showed that the optimal media was Richard and Czapek,the optimal nitrogen were potassium nitrate and sodium nitrate,the carbon were sucrose,mannitol and maltose.Indoor toxicity tests against pathogen showed that pyrazole tetracycline and fenaminosulf had a better inhibitory effect on mycelial growth,with EC50 value<1 mg·L-1,the second was Metalaxyl·Hymexazol.
