- 以兰科植物青天葵分离出的具有促生作用的菌根菌(Epulorhiza sp.1219)为试材,采用真菌平板培养法,设计10种基础培养基,并设置碳源、氮源、pH、温度等培养条件进行单因素试验,研究了青天葵菌根真菌在不同培养条件下的营养需求特性及天然产物培养基及人工合成培养基对青天葵菌根菌生长的影响,以期为瘤菌根菌属真菌的环境适应性提供参考依据。结果表明:最适宜瘤菌根菌生长的培养基为1/2CPDA培养基,配方为20%马铃薯汁500 mL·L-1+KH2PO43 g·L-1,+MgSO4·7H2O 1.5 g·L-1+葡萄糖20 g·L-1+维生素B10.5 mg·L-1+琼脂12 g·L-1,该菌根真菌在10~48 ℃时均能生长,最适温度为30 ℃,致死温度为49 ℃,可在偏酸性环境中(pH为5.0~7.0)生长;在碳源利用上,麦芽糖最佳;在氮源选择上,甘氨酸最优;以玉米芯为固体培养料时,长势最佳。综上所述,青天葵促生菌根真菌在1/2CPDA培养基、pH为6.0、30 ℃暗培养最适宜,以麦芽糖为碳源、甘氨酸为氮源,玉米芯为固体培养料时表现出最佳生长特性。
- Abstract:
- Taking the mycorrhizal fungus (Epulorhiza sp.1219) with growth-promoting effects on Nervilia fordii (Hance) Schltr.isolated from the Orchidaceae as the test material,the fungal plate culture method was adopted.Ten kinds of basic culture media were designed,and culture conditions as carbon sources,nitrogen sources,pH levels,and temperatures were set for single-factor tests.The effects of natural product media and synthetic media on the growth of the mycorrhizal fungus of N.fordii,as well as the nutrient requirement of the mycorrhizal fungus of N.fordii under different culture conditions were studied,in order to provide a reference for the environmental adaptability of the mycorrhizal fungi of Epulorhiza sp.The results showed that the most suitable culture medium for the growth of the mycorrhizal fungus was 1/2CPDA medium,consisting of 500 mL·L-1 potato juice (20%),3 g·L-1 KH2PO4,1.5 g·L-1 MgSO4·7H2O,20 g·L-1 glucose,0.5 mg·L-1 vitamin B1,and 12 g·L-1 agar.This mycorrhizal fungus could grow within the temperature range from 10 ℃ to 48 ℃,with the optimum temperature being 30 ℃ and the lethal temperature being 49 ℃.It could grow in a slightly acidic environment (pH 5.0-7.0).Maltose was the best in terms of carbon source utilization,glycine was the optimal in terms of nitrogen source selection,and the growth was the best when corncob was used as the solid culture material.In conclusion,the growth-promoting mycorrhizal fungus of N.fordii was most suitable for dark culture in 1/2CPDA medium at pH 6.0 and 30 ℃,with maltose as the carbon source,glycine as the nitrogen source,and corncob as the solid culture material,showing the best growth characteristics.
