- 以23个离褶伞属菌株为试材,采用菌丝生长形态分析和SSR分子标记技术方法,研究了种质资源遗传多样性和分子身份证的构建,以期为离褶伞属种质资源鉴定和高效利用提供参考依据。结果表明:23个菌株在PDA综合培养基上菌丝生长速度有明显差异,为1.138~2.577 mm·d-1,大部分菌株菌丝呈放射状,边缘较淡。多态性位点丰富,10对SSR引物在23个离褶属菌株中扩增得到39个多态性位点,每对引物扩增出的多态性位点数为 2~7个。供试菌株遗传相异系数在0.14~0.82,相异系数在0.54时可将菌株分为六大类群。基于10对SSR引物组合扩增出的清晰条带,构建了23个离褶伞属菌株的二维码分子身份证。
- Abstract:
- Twenty-three Lyophyllum strains were used as test materials,the genetic diversity of germplasm resources and the construction of molecular identity were studied by using the analysis of mycelial growth morphology and SSR molecular marker technology,in order to provide reference for germplasm identification and efficient utilization of Lyophyllum.The results showed that the 23 strains showed significant differences in mycelial growth rate on PDA integrated medium,ranging from 1.138 mm·d-1 to 2.577 mm·d-1,and most of the mycelium showed radial pattern with weak margins.Polymorphic loci were abundant,ten pairs of SSR primers were amplified to obtain 39 polymorphic loci in 23 Lyophyllum strains and with the number of polymorphic loci ranging from 2 to 7 for each pair of primers.The genetic phase divergence coefficient of the test strains was between 0.14 and 0.82,and when the phase divergence coefficient was at 0.54,the strains could be divided into 6 groups.The QR code molecular ID of 23 Lyophyllum strains were constructed based on clear bands amplified from 10 pairs of SSR primers.
