猴头菇凝集素的提取工艺研究

- 以猴头菇为试材，采用磷酸缓冲液（PBS）浸提法制备提取液，在浸提剂pH值、料液比、浸提时间、浸提温度等单因素试验的基础上，采用正交实验设计，研究了猴头菇凝集素的最佳提取工艺。结果表明：最佳的提取工艺以PBS（0.10 mol/L，pH 6.5）为提取剂，料液比1∶15 （g/mL），提取温度4℃，提取时间24 h。在此条件下，猴头菇凝集素的凝集活性达到0.586，提取率达到1.35%。
- Abstract:
- Taking Hericium erinaceus as material，lectin was extracted with PBS extraction method.On the basis of single factor tests of pH values of the extracting solvents，solid-liquid ratio，extraction time，extraction temperature，orthogonal experiment was designed to optimize the extraction condition of Hericium erinaceus lectin(HEL).The results showed that the extracting solvents was PBS，the concentration of sodium chloride was 0.10 mol/L.The best extraction technology was solid-liquid ratio 1∶15 （g/mL），pH values of the extracting solvents 6.5，extraction temperature 4℃,extraction time 24 hours.Under these conditions，agglutinating activity of HEL reached 0.586 and extraction rate reached 1.35%.
