- 以两面针为试材,采用响应面法,研究了回流提取时间、提取溶剂浓度和料液比对氯化两面针碱、白屈菜红碱和血根碱提取得率的影响,以期获得两面针中3种生物碱的最佳回流提取工艺,为深入开发利用两面针中的生物碱提供参考依据。结果表明:3种生物碱的最优回流提取条件为提取时间2.4 h·次-1(重复提取2次)、提取溶剂67%乙醇、料液比1∶20 g·mL-1,此时氯化两面针碱、白屈菜红碱和血根碱的得率分别为0.381%、0.560%、0.058%。该结果与响应面法预测值接近,说明该研究拟合的3种生物碱响应面回归模型可靠。优化后氯化两面针碱、白屈菜红碱和血根碱的得率分别提高了14%、95%、23%,优化工艺提取效率高,可用于两面针中3种生物碱的制备。
- Abstract:
- Taking Zanthoxylum nitidum as the experimental material,the effects of reflux extraction time,extraction solvent concentration and solid-liquid ratio on the extraction rate of nitidine chloride,chelerythrine and sanguinarine were studied by response surface method,in order to obtain the optimal reflux extraction process of the three alkaloids,and provide reference for further development and utilization of alkaloids in Zanthoxylum nitidum.The results showed that the optimal reflux extraction condition of the three alkaloidsas follows,extraction time was 2.4 hours per time (repeated twice),extraction solvent was 67% ethanol,solid-liquid ratio was 1∶20 g·mL-1,and the yield of nitidine chloride,chelerythrine and sanguinarine were 0.381%,0.560%,0.058%,respectively.The results were closed to the predicted values of response surface method,it indicated that the three alkaloids response surface regression models fitted in this study were reliable.After optimization,the yield of nitidine chloride,chelerythrine and sanguinarine were increased by 14%,95% and 23% respectively,and the optimized extraction efficiency was high,which could be used for the preparation of three alkaloids in Zanthoxylum nitidum.
