基于SRAP标记的三十三份白及种质资源聚类分析及DNA指纹图谱的构建

- 以白及(Bletilla striata Rchb.f.)为试材,基于UPGMA 法通过13对SRAP 引物组合对6个野生居群的33份白及种质资源进行聚类分析并构建DNA指纹图谱,以期为白及品种鉴定和资源收集提供参考依据。结果表明:13对SRAP 引物组合共扩增出182条条带(其中有152条为多态性条带),多态性比率为83.52%,平均Shannon′s信息指数(I)和Nei′s基因多样性指数(H)分别为0.484 6和0.324 3。6个野生居群根据UPGMA聚类,可分为2个支系。我国华南地区广东茂名、广西百色和华中地区湖南张家界3个白及居群聚为一支;西南地区贵州安龙、云南蒙自和四川峨眉山另外3个白及居群聚为另一支。Mantel test 检测表明,物种遗传距离和地理距离间存在显著正相关性 (r=0.624 6;P<0.001)。4对SRAP 核心引物组合构建了33份白及品种的DNA 指纹图谱。
- Abstract:
- A total of 33 wild Bletilla striata Rchb.f.from six different province were studied with 13 pairs of SRAP combining primers,clustering analysis and fingerprint construction of 33 germplasm resources of Bletilla striata Rchb.f.was carried out,in order to provide reference of plant resourceses protection and mixed falsify for the appraisal.The results showed that 13 SRAP combining primers produced 182 bands,included 152 polymorphic bands,with a polymorphism ratio of 83.52%.The average Shannon′s information index and the average gene diversity index were 0.484 6 and 0.324 3.Based on UPGMA,six populations gathered in two clusters.The cluster Ⅰ included three populations.Three populations were respectively taken from Guangdong province,Guangxi province and Hunan province.The another three populations which came from Sichuan province,Guizhou province and Yunan province belonged to the cluster Ⅱ.Mantel test revealed significant positive correlation between genetic distances and geographic distance (r=0.624 6;P<0.001).The DNA fingerprinted of 33 germplasm resources of Bletilla striata Rchb.f.were constructed by with four core SRAP combining primers.
