- 以“杭茄1号”紫茄子苗为试材,采用RT-PCR方法克隆CHS基因,利用DNAMAN、MEGA 7.0.26及一些在线网站研究了CHS基因所编码蛋白的结构与功能,以期为研究查尔酮合酶CHS基因在茄子的生长发育中的意义及植物类黄酮的生物合成及其分子机制提供参考依据,同时丰富了双子叶植物CHS超家族的相关研究。结果表明:获得CHS基因全长为1 170 bp,预测该基因编码的蛋白很有可能定位于细胞质中。系统进化分析表明,紫茄子与番茄、芒果、咖啡、短茄在同一个分支,亲缘关系最近,均属于茄科植物。
- Abstract:
- ‘Hang Eggplant No1’ was used as material,the CHS gene was cloned by RT-PCR,and the structure and function of the protein encoded by the CHS gene were analyzed by DNAMAN,MEGA 7.0.26 and some online websites,in order to provide reference for the study of the significance of chalcone synthase gene CHS in the growth and development of eggplant and the biosynthesis and molecular mechanism of plant flavonoids,and also enrich the related studies of CHS superfamily in dicotyledons.The results showed that the full length of CHS gene encoded protein was 1 170 bp,and it was predicted that CHS gene might be located in cytoplasm.The phylogenetic analysis showed that aubergine eggplant,tomato,mango,coffee and nightshade were in the same branch,and they all belong to nightshade family.
