- 以‘Siberia’‘Martinique’‘After Eight O′clock’‘Sunshine Iceland’和‘Bonaire’为亲本进行杂交育种,并进行正反交实验,比较正反交结实率及有胚率;采用胚挽救技术克服受精后障碍,比较不同杂交组合、胚龄、培养方式、培养基对幼胚生长萌发的影响,以期建立一套适合东方百合‘Siberia’的正反交及胚挽救方案,创制东方百合新种质。结果表明:‘Siberia’与 ‘Martinique’‘After Eight O′clock’正反交存在受精障碍,但均获得了膨大子房及有胚种子;其中,‘Siberia’作父本的有胚率显著高于其作母本,在‘Siberia’与‘After Eight O′clock’组合中,其作父本的有胚率是其作母本的4.5倍;在‘Siberia’בMartinique’组合中有胚率是其作母本的2.3倍;在‘Siberia’בBonaire’ 杂交组合中有胚率是其作母本的1.01倍;在‘Siberia’ בSunshine Iceland’ 杂交组合中有胚率是其作母本的1.2倍。胚挽救试验结果显示,70 d胚龄的幼胚最适合剥除胚乳培养,在‘Siberia’בSunshine Iceland’杂交组合中,其剥除胚乳培养的萌发率最高,是未剥除胚乳培养的5.5倍;胚离体培养的最佳培养基为MS+1.0 mg?L-1 6-BA+0.1 mg?L-1NAA+0.3%蔗糖。杂交后的种子在播种20 d左右开始萌动,萌发率在0.8%~8.7%,40 d左右出苗率显著提高,其中在‘Siberia’בBonaire’杂交组合中最高达64.3%。

- Crossbreeding was carried out by using oriental lily ‘Siberia’‘Martinique’ and ‘After Eight O′clock’ as the materials on the basis of investigating the effect of different storage duration on their pollen viability,and the reciprocal crosses were conducted to compare their fruiting rates and embryonic rates.In addition,the effects of different hybridization combinations,developing-embryo age,culture methods and media on the growth and germination of young embryos were compared through embryo rescue technology in overcoming post-fertilization obstacles,in order to construct a scheme suitable for the reciprocal crosses and embryo rescue of oriental lily ‘Siberia’ and cultivate a new germplasm of oriental lily.The results showed that there existed fertilization barriers between them,while both inflated ovaries and embryonic seeds were obtained in all of them.Among them,the embryonic rate in the hybridization combinations using ‘Siberia’ as the male was significantly higher than that in the female ‘Siberia’.In the combination of ‘Siberia’ and ‘After Eight O′clock’,the embryonic rate of using ‘Siberia’ as the male was 4.5 times that of the female,while in the ‘Siberia’בMartinique’ combination,the embryonic rate was 2.3 times that of the female.In the ‘Siberia×Bonaire’ combination,the embryonic rate was 1.01 times that of the female,in the ‘Siberia×Sunshine Iceland’ combination,the embryonic rate was 1.2 times that of the female.According to the embryo rescue results,it showed that the 70 days embryos were the most suitable for stripping endosperm culture.In the ‘Siberia’בSunshine Iceland’ hybrid combination,the germination rate of the stripped endosperm culture was the highest among them,which was 5.5 times than that of the unscathed endosperm culture.The optimal medium for embryo culture in vitro was MS+1.0 mg?L-16-BA+0.1 mg?L-1NAA+0.3% sucrose.After hybridization,the seeds began to germinate around 20 days of sowing,the germination rate was 0.8%-8.7%,and the emergence rate was significantly increased around 40 days,of which the highest in the ‘Siberia’בBonaire’ hybrid combination was as high as 64.3%.
