- 以木芙蓉品系‘单瓣红’(DBH)、‘单瓣白’(DBB)、‘牡丹粉’(MDF)和‘醉云’(ZY)以及木芙蓉品种‘金秋红’(JQH)为试验材料,在水培条件中添加NaCl模拟盐胁迫的情况下,分析了不同品种(品系)表型变化、株高、鲜质量、活性氧、丙二醛及抗氧化物酶活性,研究了不同木芙蓉品种(品系)对盐胁迫的耐受性和响应,以期为木芙蓉耐盐品种选育、抗盐机制探究及其在西北和东北地区推广提供参考依据。结果表明:在盐胁迫下,不同品种(品系)木芙蓉幼苗生长均受到抑制,NaCl处理相比CK,株高降低48.40%~58.56%,鲜质量减少63.87%~76.58%;过氧化氢(H2O2)、超氧阴离子(O?〖TX--*9〗2)积累量显著增多,叶片中丙二醛(MDA)含量增加1.65~10倍;抗氧化物酶活性明显增强,为CK的2~26倍;不同木芙蓉品种(品系)在盐胁迫处理下的耐受性表现出明显差异,‘DBH’耐受性最强,‘JQH’/‘MDF’耐受性次之,‘DBB’/‘ZY’耐受性最差。1% NaCl处理下,‘DBH’株高、根长和鲜质量为‘DBB’/‘ZY’的1.19/1.20、1.17/1.16、1.45/1.60倍;H2O2、O?〖TX--*9〗2的积累量相比‘DBB’/‘ZY’显著减少;MDA含量仅为‘DBB’/‘ZY’的73.58%/69.40%;超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)是‘DBB’/‘ZY’的1.45/1.71、2.07/1.99、1.99/2.12倍。说明盐胁迫下木芙蓉能通过提高抗氧化酶活性,减缓氧化损伤及膜损伤,缓解其生长抑制。
- Abstract:
- Cultivar (‘JQH’) and strains (‘DBH’‘DBB’‘MDF’‘ZY’) of Hibiscus mutabilis L. were used as experimental materials.The phenotypic changes,plant height,fresh weight,reactive oxygen species,malondialdehyde and antioxidant enzyme activities of different cultivars (strains) under the condition of adding NaCl to simulate salt stress under hydroponic conditions were studied,in order to explore different Hibiscus mutabilis L. cultivars (strains) tolerance and response to salt stress.The results showed that under the salt stress treatment,the growth of the seedlings of different cultivars (strains) of Hibiscus mutabilis L. was inhibited.Compared with the CK,the NaCl treatment reduced the plant height by 48.40%-58.56% and the fresh weight by 63.87%-76.58%,the accumulation of superoxide anion (O?2) and hydrogen peroxide (H2O2) increased significantly,the content of malondialdehyde (MDA) in the leaves increased by 1.65-10 times;the activity of antioxidant enzymes was significantly enhanced,which was 2-26 times that of CK.The tolerance of different Hibiscus mutabilis L. cultivars (strains) under salt stress treatment showed obvious differences,with ‘DBH’ having the strongest tolerance,‘JQH’/‘MDF’ second,and ‘DBB’/‘ZY’ having the worst tolerance.Under 1% NaCl treatment,the plant height,root length and fresh weight of ‘DBH’ were 1.19/1.20,1.17/1.16,1.45/1.60 times that of ‘DBB’/‘ZY’;compared with the accumulation of H2O2 and O?2 ‘DBB’/‘ZY’ was significantly reduced;MDA content was only 73.58%/69.40% of ‘DBB’/‘ZY’;superoxide dismutase (SOD),peroxidase (POD),catalase (CAT) was 1.45/1.71,2.07/1.99,1.99/2.12 times of ‘DBB’/‘ZY’.The results showed that Hibiscus mutabilis L. under salt stress could slow down oxidative damage and membrane damage by increasing the activity of antioxidant enzymes,thereby alleviating its growth inhibition.This research lays the foundation for the breeding of salt-tolerant cultivars (strains) of Hibiscus mutabilis L.,the exploration of salt-tolerant mechanism of Hibiscus mutabilis L. and its promotion in the northwest and northeast regions.
