商丘地区草莓叶斑病的分离与鉴定

- 以商丘双八镇草莓为试材,〖JP+2〗采用组织分离法,研究了引起草莓叶斑病的病原菌鉴定,以期为草莓种植过程中叶斑病防治提供参考依据。结果表明:引起草莓叶斑病的病原菌是一株真菌,编号为SQZQ-06,通过柯赫法则对该菌株的致病性进行测定,发现该菌株可以导致草莓叶片出现斑状病害特征。PCR扩增SQZQ-06菌株18S rRNA基因得到长度为589 bp的序列片段,将测定结果在NCBI网站进行比对并作系统进化树分析,综合该真菌的形态学特征后确定SQZQ-06为斑污拟盘多毛孢Pestalotiopsis maculans。
- Abstract:
- Taking the leaves of strawberry in Shuangba town,Shangqiu as material,the pathogen of strawberry leaf spot disease by tissue isolation was investigated,in order to provide a reference for the control of strawberry leaf spot disease.The results showed that a fungus SQZQ-06 from strawberry leaf spot in Shuangba town,Shangqiu,was identified as Pestalotiopsis maculans.The pathogenicity of the strain was determined by Coch rule,and it was found that the strain could lead to the necrosis of the strawberry leaves.The 18S rRNA gene of SQZQ-06 was amplified by PCR for molecular identification,and the length was 589 bp.Blast result in NCBI and phylogenetic tree analysis suggested that SQZQ-06 was Pestalotiopsis maculans.According to the morphological characteristics and molecular identification of the fungus,SQZQ-06 was identified as Pestalotiopsis maculans.
