色素万寿菊雄性不育植株的快速繁殖

- 以色素万寿菊为试验材料,采用母本资源不育株叶片、带腋芽茎段和侧芽为外植体,对不同的培养基配方条件下的生长情况进行统计,研究了灭菌处理、生长调节剂组合、AgNO3对丛生芽诱导的影响,以期快速获得遗传稳定的万寿菊雄性不育植株,为杂交育种提供基础研究材料。结果表明:以带腋芽的茎段为最佳外植体,1 g?L-1 HgCl2灭菌10~12 min的灭菌效果最好,B1-3较易诱导丛生芽,其最佳的诱导培养基为MS+1.0 mg?L-1 6-BA+0.5 mg?L-1 NAA+30 g?L-1蔗糖+7 g?L-1琼脂,诱导率高达93%,2.0 mg?L-1 AgNO3有利于丛生芽增殖,使B1-3丛生芽增殖率达151.6%,丛生芽生根培养后移栽成活率达95%以上。发现万寿菊离体培养过程中,外植体不同,其丛生芽诱导率不同,最佳外植体为带腋芽茎段。不同的灭菌剂的灭菌效果不同,1 g?L-1 HgCl2灭菌效果最好;丛生芽诱导时6-BA与NAA不同浓度组合优于6-BA与IAA组合;低浓度AgNO3能够促进丛生芽诱导,高浓度反而会降低。

- Pigment marigold was selected as research object.Leaves,stem with axillary buds and lateral buds of sterile plants from pigment marigold mother resources were used to research the effects of sterilization treatment,growth regulator combination and AgNO3 concentration on multiple shoots induction.The aim was to obtain the genetically stable male sterile plants of marigold,and provide basic research materials for cross breeding.The results showed that stem with axillary buds were the best explants.The best sterilization agent was 1 g?L-1 HgCl2 to sterilize 10-12 minutes.B1-3 was easy to induce multiple shoots,MS+1.0 mg?L-1 6-BA+0.5 mg?L-1 NAA+30 g?L-1 sucrose+7 g?L-1 agar was optimal induction medium,the induction rate for B1-3 was up to 93%.2.0 mg?L-1 AgNO3 was beneficial to proliferation of multiple shoots,and the proliferation rate of B1-3 was to 151.6%.After rooting culture,the survival rate of transplanting was over 95%.In vitro culture of marigold,the rate of cluster bud induction was different with different explants.Stem with axillary buds were the best explants.Different sterilization agents had different influenceson sterilization effect,1 g?L-1 HgCl2 of sterilization effect was the best.The combination of 6-BA and NAA was better than that of 6-BA and IAA for cluster buds induction.Low concentration AgNO3 could promote the induction of cluster buds,but high concentrations had opposite effect.
