- 以不同品种的红豆杉枝条为试材,利用L9(34)正交实验方法,探讨了在温室环境条件下,东北红豆杉不同插穗年龄、不同浓度ABT1以及不同浸泡时间对扦插繁殖生根率的影响,并确定了通用于其它种红豆杉扦插繁殖的最佳试验方案。结果表明:3种因素中插穗年龄和浸泡时间显著影响东北红豆杉生根率,而ABT1的浓度对东北红豆杉生根率不存在显著作用。此外,方案组合A1B3C3生根率最高,而A3B1C3生根率与之较为接近,即同为一年生枝条(C3),在较低浓度ABT1(A1:50 mg?L-1)处理较长时间(B3:4.0 h)和在较高浓度ABT1(A3:500 mg?L-1)处理较短时间(B1:0.5 h)均能够得到较高生根率(96%左右),表明较低浓度ABT1浸泡时间稍长,较高浓度ABT1浸泡时间稍短,有利于插穗生根。将A1B3C3组合在南方红豆杉和曼地亚红豆杉中进行验证,2种红豆杉扦插生根率均达到90%以上。该研究所建立的温室内扦插繁殖技术方案,简便易行,可以为其它种红豆杉扦插繁育所借鉴。
- Abstract:
- Different Taxus branches were selected as test materials,using the L9(34) orthogonal test method,9 combinations of cuttage propagation of Taxus cuspidata were carried out in greenhouse.Each combination was repeated 3 times with 50 cuttings,and the rooting rate was investigated and analyzed.The effects of the age of cuttings,the concentration of ABT1,and the length of soaking time on the rooting rate were analyzed and discussed.Moreover,the best combination for cuttings propagation of yew was obtained.The results showed that the rooting rate of T.cuspidata was significantly influenced by the age of cuttings and the length of soaking time,while the concentration of ABT1 had no significant effect.In addition,the program A1B3C3 had the highest rooting rate,and the rooting rate of A3B1C3 was nearly to A1B3C3.In details,the same one-year branch (C3),higher rooting rate could be obtained at lower concentration ABT1 (A1,50 mg?L-1) for longer time (B3,4.0 hours) or at higher concentration ABT1 (A3,500 mg?L-1) for shorter time (B1,0.5 hour) (rooting rate about 96%).The results indicated that with lower concentration ABT1,the treating time was longer,while with higher concentration ABT1,the treating time was shorter.Moreover,the combination A1B3C3 was used in Taxus chinensis var.mairei and Taxus×media,and the rooting rates of both yew reached more than 90%.The technology of cuttage propagation established by this research was simple and easy to use,and could be used for reference for other species of yew cuttage breeding in greenhouse.
