- 以霍山石斛、铁皮石斛、铜皮石斛和河南石斛根、茎、叶为试材,采用冷凝回流法、蒽酮-硫酸比色法、福林酚比色法对其水浸出物与乙醇浸出物得率、总多糖与总多酚含量进行对比,采用DPPH自由基法研究4种石斛试材清除自由基能力差异,以期为石斛资源的综合开发利用提供参考依据。结果表明:水浸出物提取得率与总多糖含量为茎>叶>根;霍山石斛、铁皮石斛与河南石斛乙醇浸出物提取得率为叶>根>茎,铜皮石斛乙醇浸出物提取得率为叶>茎>根;4种石斛总多酚含量均表现为根>叶>茎;DPPH自由基清除能力以根的效果最好。4种石斛的不同部位的水浸出物和乙醇浸出物提取得率、总多酚含量、DPPH自由基清除活性不同。以多糖含量为评价依据,茎入药较好。以总多酚的含量及DPPH自由基清除活性为评价依据,根入药较好。
- Abstract:
- The yield of the water and ethanol extracts as well as the polysaccharide content,polyphenol content and the DPPH free radicals scavenging activity from the roots,stems and leaves of Dendrobium huoshanense,Dendrobium officinale,Dendrobium moniliforme and Dendrobium henanense, were analyzed to provide references for the utilization of Dendrobiums,by condensation reflux method,anthracene-sulfuric acid colorimetric method and the colorimetric method of Folin-Ciocalteu and DPPH free radicals method,respectively.The results showed that the yield of water extract and the content of total polysaccharide were stem>leaf>root.The yield of alcohol extract of D.huoshanese,D.officinale and D.henanese were leaf>root>stem,and D.moniliforme was leaf>stem>root.The content of total polyphenol in all the four experimental Dendrobiums were root>leaf>stem;both the water and ethanol extracts from the root showed the best DPPH free radicals scavenging ability.From our investigation,it could be concluded that the content of the water extract,ethanol extract,total polysaccharide,the total polyphenol and the DPPH free radicals scavenging activity of the root,stem and leaves are quite different.Using the contents of the total polysaccharide as the quality evaluation standards,the stems of the four Dendrobiums are the most proper to use as the drug.Using the the content of the total polyphenol and the DPPH free radicals scavenging activity as the quality evaluation standards,the root might be the best medicinal parts of D.huoshanese,D.officinale,D.moniliforme and D.henanese.
