- 以菌草灵芝菌糟为试材,采用国家标准、农业行业标准等测定了提取物的粗多糖、三萜类物质、氨基酸等成分含量,并在实验室采用正交实验法对其粗多糖进行提取,研究了不同的料液比、提取温度、提取时间和提取次数对菌糟多糖得率的影响,获得最优化的提取条件,在此基础上优化了菌草灵芝菌糟提取物中试生产工艺。结果表明:菌草灵芝菌糟提取物中主要成分粗多糖含量为24.16%,三萜类物质含量为0.82%,氨基酸总量为6.50%;实验室菌草灵芝菌糟粗多糖的最佳提取条件为料液比1∶20 g?mL-1,提取温度100 ℃,提取时间4 h,提取次数3次,在最佳条件下,菌草灵芝多糖得率为4.45%;确定菌草灵芝菌糟提取物中试提取条件为提取次数3次,第1次料液比1∶10 g?mL-1,加热至100 ℃后保持2 h,第2次料液比1∶8 g?mL-1,加热至100 ℃后保持2 h;第3次料液比1∶6 g?mL-1,加热至100 ℃后保持1.5 h,平均提取得率为20.03%。

- JUNCAO-cultivated Ganoderma lucidum spent mushroom substrate was used as test material.The content of crude polysaccharides,triterpenes and amino acids and other ingredients in the water extract of Ganoderma lucidum spent mushroom substrate (SMSG-WE) was determined by using national standards,agricultural industry standards,and the extraction process of crude polysaccharides from SMSG was optimized by orthogonal test in the laboratory,the effects of different solid-liquid ratio,extraction temperature,extraction time and times on the yield of polysaccharide from Ganoderma lucidum spent mushroom substrate were studied to obtain the optimal extraction conditions.On this basis,the pilot production process of SMSG-WE was optimized.The results showed that the contents of crude polysaccharides,triterpenes and amino acids were 24.16%,0.82% and 6.50%.In laboratory the optimum extraction condition of polysaccharides was solid-liquid ratio 1∶20 g?mL-1,extracting temperature 100 ℃,extraction time 4 hours,extraction times for 3 times,under the best conditions,the polysaccharide yield is 4.45%;pilot extraction conditions of SMSG-WE were determined as follows,extraction for 3 times,for the first time the solid-liquid ratio 1∶10 g?mL-1,heated to 100 ℃ for 2 hours,second time solid-liquid ratio 1∶8 g?mL-1,heated to 100 ℃ for 2 hours,third time solid-liquid ratio 1∶6 g?mL-1,keep 1.5 hours after heated to 100 ℃,the average obtained at a rate of 20.03%.
