- 利用RAPD分子标记技术对长白山地区24个野生软枣猕猴桃种质资源进行检测,应用NTSYSpc 2.10e软件对遗传一致性和遗传距离进行分析。以期解析长白山地区野生软枣猕猴桃种质资源的遗传多样性、亲缘关系。结果表明:用24个扩增效果良好的引物进行RAPD扩增反应后,共扩增出187条带,其中多态性条带为181条,占96.8%,平均每个引物产生7.54个多态性条带。24个野生软枣猕猴桃种质资源之间遗传距离为0.02~0.58,并具有同一地理区域聚类趋势,且不同地理区域间存在较高的遗传多样性。
- Abstract:
- The aims of this study were to evaluate the genetic diversity,genetic structure and genetic relationship of wild Actinidia argute resources in Changbai Mountain area.The results showed that genetic diversity of 24 wild A.argute resources in Changbai Mountain area was investigated by RAPD techniques.The genetic identity and genetic distance were analyzed by using NTSYSpc 2.10e software.PCR with 131 random primers and screening 24 stripes which had good ploymorphism and repetitive.There were 181 ploymorphisms stripes at 96.8% in 187 bands which were augmented by RAPD reaction.Each primer produced 7.54 ploymorphisms in average.The results by UPGMA clustering analysis and RAPD reaction showed that the genetic distance among the 24 kinds of wild A.arguta was 0.02 to 0.58 and had the same geographic area clustering.And there was genetic diversity in different geographical regions.
