- 以茶树菇为研究对象,依据基因组序列,通过长程PCR扩增获得YSG和AC0007菌株线粒体中间肽酶基因(mitochondrial intermediate peptidase encoding gene,mip)。结果表明:该基因与A交配位点连锁,可以其作为A交配位点克隆的靶位点。序列分析发现,该位点的C段编码区高度保守,设计的特异性引物可以在群体中获得mip基因的保守区,另外其N端编码区的变异区域可以为A交配位点鉴定提供特异的靶标,为食用菌遗传及育种研究提供重要的基础。
- Abstract:
- Based on the genome sequence,mitochondrial intermediate peptidase encoding gene (mip) was cloned from two strains YSG and AC0007 using long-range PCR method,and the genetic characterization in Agrocybe aegerita strains population were also investigated to test the applicability of them as the molecular mark for polarity identification.The results showed that mip was co-segregated with A mating locus and therefore it could be used for the molecular for the cloning of the latter.Sequence analysis showed C terminal of MIP protein encoding region was highly conserved,and the special primer pairs were designed for this region could amplify the mip fragments in all tested strains.Additionally,the variable encoding regions in C terminal could be used as the targeting site for the A mating locus identification,and it provided an important basis for genetics and breeding of edible mushroom.
