- 以喷施砖红微杆菌Y24和解淀粉芽孢杆菌N22菌剂后的葡萄叶片为试材,采用PCR-DGGE方法研究葡萄叶片表面细菌多样性,以期阐明2种菌剂对葡萄叶片细菌多样性的影响。结果表明:经过PCR扩增出大小约为250 bp的细菌16S rDNA V3可变区。经DGGE电泳共标记出17条带,其对应葡萄叶片上的15种已知细菌包括芽孢杆菌属(Bacillus)、微球菌属(Micrococcus)、假单胞菌属(Pseudomonas)、链霉菌属(Streptomyces)等和2种非可培养细菌。其中砖红微杆菌Y24菌剂的细菌种类更为丰富,其中4条特有条带所代表的细菌类群包括微杆菌属(Microbacterium)、马杜拉放线菌属(Actinomadura)、黄假单胞菌(Pseudomonas flava)和简单芽孢杆菌(Bacillus simplex)。聚类分析表明,Y24菌剂处理后与对照及N22菌剂处理的叶片在细菌多样性方面有明显差别,而N22菌剂的处理与对照没有差别。系统发育分析表明,葡萄叶片喷施菌剂后,其表面细菌仍维持一定的多样性。砖红微杆菌Y24菌剂可显著提高葡萄叶片的细菌多样性,而解淀粉芽孢杆菌N22对葡萄叶片细菌多样性无显著性影响。

- Grape leaves that were treated by Microbacterium testaceum (Y24) and Bacillus amyloliquefaciens (N22) were taken as test materials,bacterial diversity on the surface of grape leaves was studied by PCR-DGGE to clarify the effect of two kinds of bacteria on bacterial diversity.The results showed that the size of approximately 250 bp bacterial 16S rDNA V3 variable region was amplified by PCR with total DNA extracted from the water washed grape leaves as template,17 bands were labeled by DGGE electrophoresis,those were found that 15 known bacteria on the leaves of grape,including Bacillus,Micrococcus,Pseudomonas,Streptomyces,and two uncultured bacteria.The kinds of bacteria after spraying Y24 agent were more abundant,of which four special bands represented Microbacterium,Actinomyces,Pseudomonas flava and Bacillus simplex.Cluster analysis showed that the bacterial diversity on leaves treated with Y24 was significantly different from that of the control and N22 agents,but there was no difference between the treatment of N22 and the control.Phylogenetic analysis showed that the bacterial diversity of grape leaves remained after spraying with bacterial agents.It was conclused that spraying Y24 could significantly improve the bacterial diversity of grape leaves,while N22 had no significant effect on bacterial diversity of grape leaves.
