- 以在青海省西宁市湟中县拉脊山采集的短柄龙胆(Gentiana stipitata)为试材,采用组织块分离和梯度稀释法得到一株具有抗菌活性的内生菌,分析了该内生菌的活性物质,及与龙胆属植物活性成分的关联性,以期为药用植物内生菌资源的开发利用奠定基础。结果表明:内生菌WZ-6对金黄色葡萄球菌、黄瓜褐斑病菌有明显的抑制作用,通过形态学和16S rDNA系统进化分析鉴定,WZ-6为类芽孢杆菌属(Paenibacillus),与Paenibacillus terrae的同源性最高,达到98.9%,HPLC等分析表明菌株WZ-6的发酵产物中含有环烯醚萜类抑菌活性成分。
- Abstract:
- Taking Gentiana stipitata as material.An endophytic bacterium with antibacterial activity was isolated by the method of tissue block separation and gradient dilution.Active substance of endophyte and relevance between its and active stubstance of Gentiang stipitata were analyzed,to provided foundation for the development and utilization of endophytes resources from medicinal plants.The results showed that endophytes WZ-6 had significant inhibitory effect on Staphylococcus aureus and Corynespora cassiicola.According to the characteristics of morphology and 16S rDNA phylogenetic analysis,strain WZ-6 was identified as species of genus Paenibacillus,and showed the highest similarity to Paenibacillus terrae,reaching 98.9%.The results of HPLC analysis showed that the fermented product of strain WZ-6 contained antimicrobial active component of iridoid.
