- 以大葱嫩叶为试材,采用单因素试验和正交实验探讨了Mg2+浓度、dNTPs浓度、引物浓度、Taq DNA聚合酶用量、模板DNA以及Buffer用量对大葱ISSR-PCR扩增的影响,建立了大葱ISSR反应体系。结果表明:优化后的最佳反应体系为10 μL反应体系中,10×Buffer缓冲液1.0 μL,Mg2+浓度为2.0 mmol?L-1,dNTPs浓度为0.3 mmol?L-1,引物浓度为0.65 μmol?L-1,Taq酶1.0 U,模板DNA为60 ng,用灭菌的超纯水补齐体积至10 μL。对大葱71份样品材料进行ISSR-PCR 扩增体系的检测结果显示,该优化体系扩增的产物条带清晰,稳定性高,为大葱种质ISSR分子标记遗传结构分析提供参考依据。
- Abstract:
- The young leaves of welsh onion was used as the test materials,and adopting single factor test and orthogonal test,the effect of Mg2+concentration,dNTPs dosage,primers amount,Taq DNA polymerase,template DNA and different buffer dosage on ISSR-PCR amplification of welsh onion were investigated,and the welsh onion ISSR-PCR reaction system was set up.The results showed that the optimized reaction system was established as follows,in the total reaction system with amount of 10 μL,10× buffer 1.0 μL,Mg2+ 2.0 mmol?L-1,dNTPs 0.3 mmol?L-1,primer 0.65 μmol?L-1,Taq DNA polymerase 1.0 U,template DNA 60 ng.The obtained ISSR-PCR amplification system in 71 materials was detected,and the results suggested that the bands were clear and with high stability,would provide the genetic basis for the further analysis of welsh onions.
