- 以金桂、银桂种子为试材,采用不同剂量(0、50、100、150、200、250、300 Gy)60Co-γ 射线辐射种子,观察辐射对桂花种子出苗率及幼苗生长的影响,以探讨桂花种子辐射效应问题,选育具有较高观赏价值的优良新品种。结果表明:辐射处理导致桂花相对生长率下降、种子发芽势降低、出苗时滞延长、出苗整齐度降低、根系发育受抑制等效应;辐射剂量与金桂和银桂种子的相对出苗率间呈负相关,相关系数分别为-0.86、-0.83;60Co-γ 射线辐射可引起桂花幼苗产生较多的表型变异,50 Gy处理金桂与银桂种子变异率分别达(13.6±5.23)%、(12.4±4.78)%;采用相对出苗率为指标,金桂与银桂种子辐射半致死剂量分别为92.39、73.63 Gy,银桂种子辐射敏感性高于金桂种子。
- Abstract:
- In order to study the radiative mutation breeding of Osmanthus fragrans,explore the radiation effects of Osmanthus fragrans and select new variety of Osmanthus fragrans that having high ornamental value,the Osmanthus fragrans seeds from two different varieties(var.thunbergii Makino and var.latifolius Makino) were treated with different dosages to 60Co-γ ray (0,50,100,150,200,250 and 300 Gy),and the seed germination rate and seedling growth were observed.The results showed that 60Co-γ radiation caused Osmanthus seedling relative growth rate decreased,the seeds germination potential was reduced,delays extend emergence,emergence uniformity was reduced,root development were inhibiting.The seeds germination rate of Osmanthus fragrans had a high negative correlation between radiation dose,the correlation coefficient was -0.86,-0.83.60Co-γ radiation could cause the leaf of Osmanthus produce more phenotypic variation,the variation rate of var..thunbergii Makino and var. latifolius Makino were (13.6±5.23)%,(12.4±4.78)% with 50 Gy treatment respectively.Using relative germination rate as an indicator,LD50 doses of 60Co-γ radiation for the seeds of var.thunbergii Makino and var. latifolius Makino were 92.39 Gy and 73.63 Gy,respectively.The radiation sensitivity of var.thunbergii Makino seeds was higher than var.latifolius Makino seeds.
