金钗石斛叶艺性状保持及丛生芽增殖研究

- 以金钗石斛稳定的叶艺个体(植物嵌合体)茎段作为外植体,1/2MS为基本培养基,采用单因素试验研究了不同植物生长调节剂及浓度水平对茎段腋芽处的丛生芽增殖效果。结果表明:通过茎段增殖方式,各处理均能较好的保持丛生芽的叶艺性状,保持率均在87.2%以上;植物生长调节剂的不同处理水平对金钗石斛腋芽处丛生芽的增殖均有不同程度的促进作用。该试验中,0.1 mg?L-1 TDZ较适合金钗石斛茎段丛生芽的诱导;6-BA较KT、TDZ更适合金钗石斛茎段丛生芽的生长培养,其中2.0 mg?L-1的6-BA效果最好。
- Abstract:
- The stem segment of Dendrobium nobile with axillary buds were used as explants and cultured on 1/2MS medium.Effects of the different concentration of different plant growth regulators on the propagation of the buds were studied by the single factor experimental.The results showed that each treatment could better keep the plant be chimeras traits by propagation through the stem,keeping rates was above 87.2%;all treatments promoted propagation of multiple shoots to some degree.0.1 mg?L-1 of TDZ had a significant impact on induction of multiple shoots.6-BA in comparison with KT and TDZ was more suitable for growth of multiple shoots of Dendrobium nobile and 2.0 mg?L-1 6-BA was recommendable for bud growing in this experiment.
