- 以忍冬属不同来源的4种种质为试材,采用形态分类学的方法,根据株高、叶长、叶宽、叶的形状、花开时间等农艺性状,对不同种质进行了比较分析,同时采用分子生物学的方法,以5对引物对4份种质的DNA条形码序列进行PCR扩增和测序,构建聚类图,并分析物种的亲缘关系。结果表明:忍冬属4种种质在农艺学性状上差异不明显,如采用形态学的方法进行鉴别容易导致误判,然而这4种种质在DNA条形码序列上存在着丰富的遗传多样性,研究认为在对金银花种质进行亲缘关系鉴定时,借助DNA条形码序列特性与农艺学性状相结合能有效区分物种之间的差异,鉴定结果相对可靠。
- Abstract:
- Taking four species of Lonicera as test materials,the agronomical characters of four species of Lonicera,such as the height of plant,leaf length,leaf width,leaf shape and flowering period were studied.The results showed that character difference of four species was not obvious,which easily caused misjudgment in species identification.At the same time,the genomic DNA was analyzed by agarose gel electrophoresis and PCR amplification of DNA barcode was carried out for further comparison with five pairs of primers.The phylogenetic relationship of the four species by DNA barcoding sequencing was analyzed,and the phylogenetic dendrogram was built by MEGA.The results showed that there was no obvious difference between the agronomy traits of these four species,which easily caused misjudgment in species identification.On the basis of agronomical characters and DNA barcoding identification technology,the genetic diversity of the four species was analyzed,the result showed the dentification of DNA barcoding and the comparison of agronomical characters should be considered together when identifying the genetic relationship of species.
