异质生境的新疆野扁桃组培快繁体系比较研究

- 以分布在新疆5个异质生境的野扁桃一年生初展叶茎尖为试材,采用组织培养快速繁殖的方法,研究不同生境对野扁桃居群再生体系的影响。将外植体接种于MS+6-BA 0.5 mg/L+NAA 0.3 mg/L的初代诱导培养基上,将初代诱导的无菌茎尖接种于MS基本培养基上并附加不同种类的激素及浓度的配比、不同浓度的蔗糖,比较研究5个异质生境新疆野扁桃茎尖的组培快繁体系。结果表明:芽苗增殖的最适培养基分别是“哈巴河”:MS+6-BA 1.5 mg/L+IBA 0.1 mg/L+GA3 0.5 mg/L,“布尔津”:MS+6-BA 1.0 mg/L+NAA 0.1 mg/L+GA3 0.5 mg/L,“托里”:MS+6-BA 0.8 mg/L+IBA 0.2 mg/L+GA3 1.0 mg/L,“裕民”:MS+6-BA 0.6 mg/L+IBA 0.2 mg/L+GA3 1.0 mg/L,“塔城”:MS+6-BA 1.0 mg/L+IBA 0.1 mg/L+GA3 0.5 mg/L。对增殖情况较好的“哈巴河”野扁桃芽苗进行了进一步的最佳生根培养基的筛选,“哈巴河”野扁桃芽苗最适生根培养基为1/2MS+6-BA 0.04 mg/L+IBA 1.5 mg/L。
- Abstract:
- The test materials were annual exhibition of the stem tips of Prunus tenella in five heterogeneous habitat of Xinjiang,by adopting the method of tissue culture of rapid propagation,the effect of populations in different habitats of Prunus tenella regeneration system was studied,and the primary explants were inoculated on the induction medium,which was composed of MS+6-BA 0.5 mg/L+NAA 0.3 mg/L,then inoculated in MS basic medium and different hormone,additional different concentration,different sucrose concentration.It was discussed that shoot spex proliferation ability on five heterogeneous habitat distribution populations of wild almonds.The results showed that the most suitable medium seedling proliferations were‘Habahe’:MS+6-BA 1.5 mg/L+IBA 0.1 mg/L+GA3 0.5 mg/L,‘Buerjin’:MS+6-BA 1.0 mg/L+NAA 0.1 mg/L+GA3 0.5 mg/L,‘Tuoli’:MS+6-BA 0.8 mg/L+IBA 0.2 mg/L+GA3 1.0 mg/L,‘Yumin’:MS+6-BA 0.6 mg/L+IBA 0.2 mg/L+GA3 1.0 mg/L,‘Tacheng’:MS+6-BA 1.0 mg/L+IBA 0.1 mg/L+GA3 0.5 mg/L.The optimum rooting culture medium of ‘Habahe’ whose proliferation was better was screened.The optimal rooting medium of ‘Habahe’ materials was 1/2MS+6-BA 0.04 mg/L+IBA 1.5 mg/L.
